|
This study was carried out to investigate several factors affecting RAPD band patterns in apple and to establish the optimum RAPD condition for apple. The results are as follows: The optimum composition of PCR reaction solution for RAPD assay of Malus domestica was 10 ng template DNA, 2 units Tag polymerise, 10 ng primer, 50∼100 μM dNTP, 2.5 ㎕ 10X reaction buffer and fill up to 25 ㎕ with sterile distilled water. Fifty cycles of amplification produced best RAPD band patterns.
