Factors Influencing RAPD Band Patterns in Malus domestica

Abstract
This study was carried out to investigate several factors affecting RAPD band patterns in apple and to establish the optimum RAPD condition for apple. The results are as follows: The optimum composition of PCR reaction solution for RAPD assay of Malus domestica was 10 ng template DNA, 2 units Tag polymerise, 10 ng primer, 50¡­100 ¥ìM dNTP, 2.5 §¡ 10X reaction buffer and fill up to 25 §¡ with sterile distilled water. Fifty cycles of amplification produced best RAPD band patterns.
